Production of cortical organoids as a model for brain development

The brain is a particularly complex organ both in terms of structure and connectivity. It is formed during development by the ordered and sequential differentiation of stem cells that organize and differentiate into a three-dimensional (3D) lamellar structure. Cellular divisions of progenitors in the external sub-ventricular area (OSVZ) of the cortex ensure its expansion. A patent application text has been filed describing an invention that aims to reproduce in vitro in 3D structure the organization, cellularity and connectivity of this cortical zone by producing different differentiated and organized cellular types. The organoids produced at the end of this process mimic the cellular behaviors observed in vivo and allow the testing of many molecules of interest potentially involved in infectious, neurodegenerative or behavioural pathologies.
